According to recent estimates approximately 60 percent of the heritability of LOAD has been found. The next challenge will be to understand mechanistically how these genes influence downstream pathology. As for the 'missing heritability', it is debatable how much more we will learn for discovering risk alleles imparting vanishingly small effects. Nevertheless, currently the PERADES consortium is assembling the largest LOAD case control series, enabling a mega GWAS in the years to come. Irrespective of the effects size, a risk-allele is an opportunity to understand the disease.  In that sense  GWAS is not should be viewed as  the end, its the start. starting point.
