In this work we build a foundation towards developing a framework for a comprehensive modeling and simulation of longitudinal structural MRIs of AD patients.  One of the immediate benefits of this framework is in evaluating and validating large number of atrophy/volume measurement algorithms existing in the literature.  Large database of ground truth synthetic structural MRIs that can be generated from the proposed framework could also be used to train machine learning algorithms that intend to compute local volume changes.  In the long run, we hope that researchers will build upon this foundational framework to develop more complex models for predicting and simulating patient-specific structural changes in the brain with AD. This sort of model that can reliably predict the trajectories of AD could be extremely useful in monitoring the clinical trials of disease modifying drugs.
